I just got my 350 v8 rebuilt for my 47 Chevy business coupe, it's fully upgraded with a stroker kit and a ford 9" rear end. But I'm debating which automatic transmission to buy to make it a comfortable cruiser. Any suggestions will help

What would the difference be between a 400 and a 700r4? also have an extra 350 trans, but guessing it would not be best for this project?

The 700r4 has overdrive (4-speed auto) which is better on gas for your cruiser...

Depends....what rearend gears does it have? how much cam? How do you define "comfortable"? Is cost an issue?
