If you define hot rod in the traditional sense, as a car based on an early Ford body and frame, then Tony Waters was the first person to go 200mph in a '32 Ford Roadster in 1958. RIP Tony Waters.
Auto-powered it would be the Kenz & Leslie 777 liner (dual engine) or the Xydias (single engine) SoCal Streamliner.
If I'm not totally off base I think the Ford GT 40 was the first American car to see 200 MPH in 1966. HRP

Buddy Baker went over 200 mph in a Dodge Daytona Charger test mule in 1969. Most of the Chrysler Daytona's, Plymouth Superbird's, Ford Torino Talladega's and Mercury Cyclone Spoilers were all capable of over 200 mph on the big tracks.

There seems to be some controversy as to what an "American hot rod" is as well. So, are you asking about the first car to achieve 200 mph in the quarter mile? Or the first car built in America to reach 200 mph? Or the first HOT ROD to reach 200 mph. If its the latter, then I already answered your question. Just sayin'.
In 1952 that WAS a hot rod. In 2013 it still IS a hot rod. It just may not fit into what is defined here as a traditional hot rod.
Flip this could go all over the place, if including land speed record car, there were cars going 200mph way way way early. Depends on whether you mean race car or road car and even then what kind of race car and a stock road car or a modified (hot rodded) road car.
